Small Dogs Tend to Live Longer Than Big Dogs. Scientists Say They Might Finally Know Why—and It Goes to the Molecular Level

Larger breeds like Great Danes seem to accumulate changes to chemical tags on their DNA more quickly than tinier breeds, such as chihuahuas. These alterations allow potentially harmful ‘jumping genes’ to move around their genomes

A Great Dane is lucky to see its 10th birthday, while a Chihuahua might live to celebrate its 20th. But exactly why smaller dogs tend to live longer than their larger counterparts has long been a mystery.

Now, new research suggests the differences may stem, in part, from what’s happening at the molecular level. A study published October 8 in the journal Science suggests big dogs’ cells age faster because of changes that allow potentially harmful “jumping genes” to move around their genomes, or complete sets of genetic instructions.

The work is “fascinating,” Borbála Turcsán, an animal behavior researcher at Eötvös Loránd University in Hungary who was not involved with the study, tells Nature’s Heidi Ledford. “It provides a molecular background for something we also observe at a behavioral level.”

Researchers behind the new study used data from the Dog Aging Project, a citizen science initiative in which participating dog owners and their veterinarians regularly share information about their pup’s health, behavior, environment and physical activity. Founded in 2014, the project has since enrolled roughly 55,000 canine companions of various breeds.

Scientists analyzed the DNA of 894 pet dogs spanning diverse breeds and body sizes. More specifically, they looked at methyl groups, or chemical tags on DNA that scientists often use as biological markers of aging. Methyl groups are part of the epigenome, chemical changes that regulate gene activity—controlling which genes are turned on or off—without affecting the underlying genetic code. Factors like diet, exercise, stress and pollutants can alter the epigenome.

They also looked at detailed demographic and genetic information. Using correlations between DNA methylation patterns and chronological age, they developed a molecular clock that could predict a dog’s age with a median error of less than one year based on its epigenome.

Analyses revealed that larger dogs tend to accumulate epigenetic changes associated with aging more quickly than small dogs, and that male dogs also tend to accrue these modifications faster than female dogs, even if they were neutered at a young age. In other words, both big dogs and male dogs showed signs of accelerated molecular aging. The study also found that dogs whose biological age was older than their chronological age were at greater risk of early death.

In large dogs, this faster molecular aging trajectory seems to be related to “jumping genes,” also known as transposable elements, or DNA sequences that can move from one location in a genome to another. These rogue sequences can interfere with normal gene function, leading to a variety of health problems, as well as inflammation and aging.

Methyl groups help keep jumping genes in place. The study suggests that large dogs tend to lose these methyl groups more rapidly as they age, which might make it easier for jumping genes to hop around and disrupt gene function.

“Large dogs show a faster-moving DNA methylation aging signature in blood, and that this acceleration is especially strong in parts of the genome that normally help keep ‘jumping genes’ quiet,” study co-author Noah Snyder-Mackler, a genomicist at Arizona State University, tells Reuters’ Marta Serafinko. In such canines, “methylation at those regions appears to erode more quickly with age, as if the genome is losing some of the tape or clamps that keep these mobile elements suppressed.”

Humans may have inadvertently sent big dogs down their accelerated molecular aging path, Snyder-Mackler adds. By breeding dogs to grow and reach their large adult sizes quickly, we may have unknowingly shortened their life spans.

“Their bodies have to make this trade-off between really rapid growth and maintenance of that, versus investment in the immune system and integrity of the organism,” Snyder-Mackler tells Live Science’s R.J. Mackenzie, adding that, for now, this is just a hypothesis.

In male dogs, meanwhile, the study suggests epigenetic changes on the X sex chromosome might be driving their accelerated molecular aging. Male dogs typically have one copy of the X chromosome, while female dogs usually have two—and the researchers think the extra copy might help female dogs stave off some of these aging-related epigenetic changes.

“This was an unexpected result,” study co-author Brianah McCoy, a biologist now at the University of North Carolina at Chapel Hill who conducted the research at Arizona State University, says in a statement. “It challenges some of our assumptions about how the X chromosome is regulated and highlights the complexity of epigenetic aging.”

The molecular aging differences between males and females appear to be distinct from the aging-related differences between big dogs and small dogs, so Snyder-Mackler considers it “a separate biology that needs follow-up, rather than the same mechanism in disguise,” he tells Reuters.

Additionally, while the study finds an association between DNA methylation patterns and longevity in dogs, it doesn’t establish whether or how these patterns contribute to aging. Researchers hope to use this paper as a jumping-off point to learn more about the relationship between DNA methylation and aging in canines.

Moving forward, the findings might one day lead to developments that could help extend the life spans of larger dogs. There “may be processes we can measure, follow over time and potentially modify before disease appears,” Snyder-Mackler tells Reuters. However, even if scientists find a way to slow down molecular aging, it wouldn’t be a cure-all. Large dogs would still face breed-specific risks that might affect their longevity, Snyder-Mackler notes.

Longer term, the research could even have implications for human aging. In the meantime, however, methylation changes are probably more like wrinkles—evidence that someone is getting older, but not an underlying cause of aging.

“Wrinkles are certainly diagnostic of aging,” Trey Ideker, a biologist at the University of Oxford in England who was not involved with the study, tells Nature. “There’s a big business in curing wrinkles. But is that going to cure aging? Probably not.”
